Due to the recent outbreak of avian influenza, transportation of frozen canine semen with egg yolk has been sharply restricted. Thus, there is urgent need to develop a novel egg yolk-free extender for freezing canine spermatozoa. In the present study, the effect of using skim milk/glucose (SG)-based extender without egg yolk on the motility and fertilizing capacity of canine spermatozoa frozen-thawed in the presence of glycerol was examined. ABSTRACT Date palm (Phoenix dactylifera L.) is a fruit tree resilient to adverse climatic conditions predominating in hot arid regions of the Middle East and North Africa. Low temperature tolerance was determined for gametophytes of food alga Undaria pinnatifida according to various cryopreservation methods. Survival durations after storage in a medium containing 28% glycerol at −80 °C or in a 5-10% glycerol solution at −196 °C were only 9 and 4 days respectively. On the other hand, cooling at 5 °C/min. and storage at −15 °C for 15 minutes, without cryoprotective medium, did not reduce viability but this was highly decreased by storage for 15 minutes at −30 or −196 °C, with or without DMSO. The morphological cooling injuries were depigmentation and/or a contraction of the cells.

The objective of this study was to analyze interactions between different straw sizes and thawing rates on the post-thaw goat semen parameters. Twenty-one ejaculates (seven per animal) were collected from three stud bucks by using an artificial vagina. After evaluation, the semen was extended in Tris-egg yolk-glycerol and packed in 0.25 and 0.50 mL straws, followed by storage in liquid nitrogen. Short-term storage and cryopreservation of turbot (Scophthalmus maximus) sperm ArchiMer

Short-term storage over several days as well as cryopreservation of turbot (Scophthalmus maximus) sperm were studied. Two extenders, Ringer 200 and artificial seminal liquid (ASL2), are suggested for semen collection in order to avoid the deleterious effect of urine contamination, and for the purpose of short-term storage between 0 and 15 degrees C. Oxygen atmosphere is not suitable for turbot sperm storage. Turbot spermatozoa undergo cryopreservation with a high rate of success especially in a sucrose solution with 10% dimethyl sulfoxide (DMSO) and 10% egg yolk.

Effects of cryoprotectants and low temperatures on hatching and abnormal embryo development of Prochilodus lineatus (Characiformes: Prochilodontidae) Neotropical Ichthyology

ABSTRACT This study evaluated the effect of the cryoprotectants and the low temperatures on the embryonic development of Prochilodus lineatus, describing their main morphological alterations. On chilling sensitivity test, the survival rates at the twenty somites stage (20S) were 53.6% at 0ºC, and 100% in 5ºC. To test toxicity, the embryos were exposed to a graded series of 1,2-Propanediol (PROP), dimethyl sulfoxide (Me2SO4) and glycerol (GLY), terminating in a solution of high osmolarity. There was no significant difference in the embryos survival of toxicity test between series of PROP and Me2SO4 in the 6S and 20S. We evaluated the effect of time and temperature on acrosin release from the acrosomal cap and the activity of this enzyme during in vitro capacitation in fresh and frozen/thawed dog sperm. Sperm-rich fractions of six ejaculates from three dogs were processed as fresh and frozen samples. Each sperm sample was incubated in canine capacitation medium (CCM) for 0, 1, 2 and 3 h at 20°C and at 37°C. This study aimed to establish a cryopreservation protocol for embryogenic cultures of A. angustifolia, enabling the ex situ conservation of the species. Embryogenic cultures were established from immature seeds and treated with variations of the cryoprotectant solutions SuDG, SoD and PVS2 prior to immersion in liquid nitrogen. Cell viability was evaluated after 30, 60 and 90 days of re-growth. The highest re-growth without morphological alterations and with normal biochemical composition was obtained with the PVS2 solution with 40 min immersion in ethanol (-20 °C). This procedure opens new horizons for the ex situ conservation of the species genetic.

Cryopreservation of sea bass (Dicentrarchus labrax) spermatozoa in experimental and production simulating conditions ArchiMer

A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. Viability of dourado embryos cooled in different cryoprotectant solutions PAB

The objective of this work was to evaluate the effect of different cryoprotectants on the viability of dourado (Salminus brasiliensis) embryos. Ten cryoprotectant solutions were tested. For each solution, 300 embryos were selected at the closing of the blastopore stage, and 300 more embryos were used as a negative control. After cooling (-8ºC for 6 hours), the embryos were rehydrated directly in the incubator until hatching. The best result is obtained with the cryoprotectant solution containing 9% methanol associated with 17% sucrose, resulting in a larvae hatching rate of 67.06%.

Ultrastructure of fresh and post thawed sperm of pejerrey Odontesthes bonariensis (Atheriniformes) Neotropical Ichthyology

In the present study it was showed for the first time the ultrastructural morphology of O. bonariensis sperm using electron microscopy techniques. Different kinds of abnormalities were described in fresh and post thawed sperm caused by crogenic protocols.
